Permanganate bleach to remove melanin
(Use poly-l-lysine coated slides.)
- Dewax sections, rinse in alcohol, rinse in water.
- Bleach in 0.25% potassium permanganate - 20 minutes.
- Rinse in distilled water.
- Decolourise in 1% oxalic acid - 1 minute.
- Rinse in distilled water.
- Either:
- Continue with another technique.
- Or counterstain with nuclear fast red - 1 minute.
- Rinse in distilled water.
- Dehydrate, clear & mount.
