Histological Staining Techniques

Goodpasture's carbol fuchsin stain for encephalitozoa
Goodpasture-Perrin 1943.
Use a positive control section.

Fix in Zenker or Orth, or formalin fix and post chrome with 2.5% potassium dichromate for 2 days prior to processing.
  • Dewax sections, rinse in alcohol, rinse in water.
  • If  Zenker fixed, remove mercury pigment.
  • Stain on a rack with carbol ailine fuchsin, heat to steaming with a flame twice over 5 minutes.
  • Rinse quickly with tap water.
  • Decolourise with 38% analar formaldehyde in a sealed coplin jar until no more colour is removed - 45 minutes.
  • Rinse with tap water.
  • Counterstain with saturated aqueous picric acid for 1 minute.
  • Dehydrate, clear and mount.


Encephalitozoa - blue/purple
Toxoplasma chromatin - brown/red
Influenza virus - blue
Nuclei - light red
Cytoplasm - pink/yellow
Erythrocytes - brown/yellow

Carbol aniline fuchsin
Basic fuchsin 0.59 g
30% alcohol 100 mL
Analine 1 mL
Phenol 1 g
