Histological Staining Techniques

Macchiavello for rickettsieae
Macchiavello, 1937.
Use a positive control.
  • Take to water
  • Stain in freshly prepared 0.25% *basic fuchsin in pH7.2 phosphate buffer - 30 minutes
  • Differentiate in 0.5% citric acid - 30 seconds to 1 minute or more (check microscopically)
  • Counterstain in 1% aqueous methylene blue - 15 to 30 seconds
  • Wash in water
  • Dehydrate, clear and mount.

Rickettsieae - red
Certain viral inclusions - red
Background - blue
